I have incorporated Silver nanoparticles into PMMA polymers and made discs with 10X2 mm and attempted to evaluate antifungal and antimicriobial properties using DIsk diffusion and Broth dilution methods. But no antibacterial properties were shown by the tested specimens. the same nanoparticles were incorporated into other materials and checked the antibacterial properties using disk diffusion method. The tested specimens showed good antifungal and antimicrobial properties.
Only in the polymers they are not showing any antibacterial properties. Kindly suggest a suitable method for evaluating the antibacterial properties of various nanoparticles (for example, silver, ZnO, CuO, TiO2 NPs) incorporated into polymers.
From your experience, we can suppose that silver particles have reacted with your polymer, so the antimicrobial effect disappeared.
You should get informations on your polymer and possible reactions that can undergone.
Try to change the polymer and/or the nanoparticles.
It seems that mixing with polymers have changed the antimicrobial effect of your nanoparticles. Kindly check the antimicrobial effect of nanoparticles alone in nutrient broth and check MICs. If the polymer has changed the pharmacological effect then change in the polymer or designing the nanoparticles in the form of liquid dosage form like suspension, semisolid dosage for like creams might be the options.
