I seek to establish certain cultivars of Malus spp. into tissue culture. I am looking to determine the optimal time to leave a specific tissue in a combination of baths (75% ethanol and 0.5% sodium hypochlorite and tween 20). I will be using the same type of tissue, and the same cultivar.

My goal is to establish the parameters within which to seek the optimally effective bath times. These are effectively pretests.

I am proposing to leave the tissue in each bath to determine the time needed to cause fatal damage to the tissue. Once that is established, I am proposing to use half that time in said bath and determining the time needed in the other bath to cause fatal damage; testing this twice, alternating the order of the baths.

Is this the right approach? Is using 50% of lethal exposure time correct? Is there a better path forward?
