What system is it prefereble to use for determination of docetaxel LD50? I utilized dissolution system that is used for clinical application, but it is not sutible in my case because of little volume of dilluent, that I should use. I also changed pH of docetaxel water solution, but it has changed collour and charasteric peak obtained by spectrophotometer. So, is there another way to increase solubility docetaxel in water for in vivo assay?
You can first dissolve docetaxel, and make a stock solution using any suitable solvents or oil. From the stock solution, take desired volume of stock solution and dilute up to respective volume with dissolution fluid. In this way, you can make solution for In vivo.
Check the solubility of Dacetaxel in any organic solvent and add small amount of it into 2mL highly soluble organic solvent and then make up to 100mL water and check the abs.
You can use cosolvent (ethanol, glycerol, propyleneglycol, DMSO,...) or micellar solubilization (cremophor, polysorbate, poloxamer, macrogol-15HS...), or try cyclodextrins but each of these technics can change the PK of the drugs, (for in vivo study).

Pradum Pundlikrao Ige, Vipul Patel, I used stock solution in DMSO (200 mkl) and dilluted it up to 3 ml with saline (concentration was about 1,6 mg/ml). Unfortunately, docetaxel was precipitated. As I need to determine LD50, I should dissolve docetaxel in high concentrations (about 3 mg/ml) and, hence, cannot use big volumes of diluents.
Frédéric Marçon, I used 95% ethanol (13% v/v) and Tween 80 (25% w/v) and added saline, but docetaxel was precipitated at this conditions.
It's obvious because water acts as an anti-solvent in DMSO and ethanol. I am not much aware about the solubility of Docettaxel but I would try stock solution in PEG or IPA alone or with Tween 80 and then dilution with saline.
