I would like to establish a soil microcosm for studying the effect of a microbial enzyme on gas exchange. How can I nullify the effect of other micro flora contributing to the microcosm? If I add any inhibitory substance like formalin or antibiotics, again that may inhibit my pure bacterial culture. Moreover, I understand that sterilisation of the soil may change the actual nature of microcosm and its gas exchange pattern. Kindly suggest any reliable method for resolving this issue.

It's Tricky business and deadlock, I don't understand the rational behind not putting your culture in soil without sterilization because its ultimate application will be in the nature/soil.
However I suggest take autoclave & non-sterile soil, inoculate your desire bacteria and check the soil respiration (its easy was to see the respond of bacteria in soil: Refer thesis entitle: " Phytate mineralization by native soil bacteria and phytase overexpressing rhizobacteria in relation to P availability for plants "). If you get more respiration in your inoculated one means your bacteria do response/survive.

Hi Vinod! I heard once of a similar experiment where the soil was treated radioactively, but maybe UV light can also do it?
