Hello,

I am having difficulty in performing the spore staining procedure. I followed the Schaeffer-Fulton method with some modifications according to other people's research. I have done three different methods, as follows:

a. Heated over the mouth of a glass beaker for 3 minutes using a culture age of 24 hours.

b. Heated over a bunsen burner and swung for 1 minute using a 48-hour culture age.

c. Heated over a bunsen and rocked for 10 minutes using a 72 hour culture age.

However, I did not find any spores, only vegetative cells. I don't know which step was wrong or whether the culture age used was inappropriate.

Could you please tell me the error of my method?
