I am aiming to isolate primary cells from freshly resected tumor tissue from consented patient and injecting those cells to mice. We have failed two times doing so, because the cells were contaminated.
My questions are ( which medium to grow the cells, suspension or attached cells? ) how to transport the cells or tissue. ( AQIX solution previously we used). And do you add antibiotics to it?

Шановна Ануша! Ми маємо великий досвід в культивуванні пухлин головного мозку людей. В наших роботах описані такі методики. Фрагмент пухлини спочатку достатньо розмістити в стерильний фізиологічний розчин (для транспортування). Потім фрагмент промити декілька раз в свіжих порціях фізиологічного розчину. Потім очистити фрагмент тканини від некротичних ділянок, обламків судин. Потім перемістити тканину в розчин СМF (без іонів Ca та Mg) посікти на маленькі шматочки. Піпеткою Пастера дісоціювати тканину. Потім клітини осадити центрифугуванням. Осад розвести в середовищі 199 (пухлини з операційної) або DMEM High Glucose (штамми пухлин) (Biowest). Якщо працювати максимально стерильно, можна обійтись і без антибіотиків. Потім клітини підраховуються і розсіваються в необхідній кількості. В наших роботах методика описана. Pasteur pipette dissociate the fabric. Then the cells were precipitated by centrifugation. Dilute the precipitate in medium 199 (tumors with surgery) or DMEM High Glucose (tumor strains) (Biowest).
