How to do Live/Dead assay of Cell-embedded Gelatin-Alginate scaffold using confocal microscopy?

I am facing problem with calcein staining as only dead cells are visible during confocal. I could clearly see cell differentiation in phase contrast microscope but at time of LD assay not a single live cell is visible. I am suspecting calcein dye is not penetrating the scaffold. Can anyone troubleshoot the problem behind this and please suggest alternate methods.
