Generally speaking, smaller volumes are preferred – you can always dilute for downstream applications but concentrating EV suspensions is a pain.
That being said, it also depends on the tube you used for UC. For smaller rotors/tubes, you might be able to use just 30 – 50 ul – for bigger tubes such as yours, you’ll probably need a larger volume just to cover the bottom of the tube appropriately.
Another factor that plays into this is the initial biofluid: EV-rich biofluids will result in pellets with lots of EVs, which means you can get away with larger volumes and still end up with a decent EV concentration. Low-yield biofluids will do better with smaller volumes for resuspension – larger volumes would just result in low-concentration EV samples, which can be tricky for some applications.
