Hello. I would like to dissolve Amphotericin B for PO admition, so I have to use non toxic solvent. This drug is lipophilic and insoluble in water. I couldn't find online how to dissolve it. Hope anybody here can help me. Thank you in advance!
Amphotericin B is insoluble in water at pH 6 to 7, but soluble in water at pH 2 or 11. It is soluble in DMSO (30–40 mg/ml) and in dimethylformamide (2–4 mg/ml). Aqueous solutions cannot be sterile filtered due to poor solubility.
Therefore, you have two choices: (1)-Dissolve it in buffers pH 2 or 11, (2)-dissolve it in DMSO (this is non-toxic solvent).
It is used without consequences in concentrations up to 70%. Therefore you might use a mixture of buffer 11/DMSO: 50/50 or Buffer 2/DMSO: 50/50 or 70%DMSO and /30% one of the two buffers.
