In currently, I'm training to design an animal model for studying DPP-4 inhibitors.
But, some problems confuse me that how to determine the dosage of in vivo study?
Or, is there any suitable cell line for MTT assy or another way for determining the dosage?

You might have to decide which route you will apply the drugs and and the optimize the dosage for the parameter(s) you want to monitor.

Thank you for introducing this article, it's helpful to me for making my research more complete. However, I still have some questions that the article seems didn't mention how to determine the dosage. Or, maybe there is some information I still don't understand fully...
I mean, I still can not get the point that how to determine the dosage in the animal model if I'm developing a new compound.

Thank you for introducing this article, it's helpful to me for making my research more complete. And I have another question, in this article, the author used different route and dosage in tested animal, and determined it according to the result. I think it would be the most direct and useful way. But, is there any other way could be used without using animal?

if you can measure DPP4 activity directly and want to characterize DDP4-inhibitors, possibly novel molecules, basically every cell line should work, as DDP-4 is quite ubiquitously expressed (see e.g. https://www.frontiersin.org/articles/10.3389/fimmu.2015.00386/full). If you're after systemic effects of e.g. incretins, you might need an animal model, though.
