We could get many mutants when we transform our target gene into the algae genome for overexpression or gene silencing .But how to check the mutant has overexpressed?By qPCR or Western Blotting? How to choose the time point for taking sample? In log-phase or stage-phase? Or both of it?

Thank you for your supplementary data! Actually, I faced this kind of case: the target gene of mutant was overexpressed when I analyzed the log-phase sample but for the stationary phase sample or ,for instance, one taken under nitrogen depleted condition, their expression was not. So is this kind of clone overexpressed tansformant?
