I would like to know how I can get a standardized suspension of Aspergillus brasiliensis. I used a paper Whatman No. 4 to separate hyphae and spores when I check in plate count at various dilutions get inconsistent results after incubating at 20 ° C to 25 ° C in three days.
Count after 5-7 days of incubation and don't disturb the plates in between, some spores may fall and give extra counts. Do proper mixing of sample with the dilluent (Use vortex-er or mix thoroughly using pipette) before plating and mix the sample completely and evenly with the media after pouring.

first and the foremost thing is that you do spore count (microscopically using a suitable counting chamber) of you culture in buffer after filtering it. It is very difficult to do the spore count by the growth on a solid medium as the newly developed spores detaches from the sporangia and falls on the same medium to give crazy results.
