Classical Silica TLC Plates (TLC)

Silica gel is the universal adsorbent used in TLC. It allows to carry out almost every type of separation by suitable choice of the mobile phase. EMD classical silica TLC plates are based on a combination of EMD silica gel 60 and the addition of a unique polymeric binder resulting in a very adherent and hard surface that will not crack or blister and even allow writing with a pencil on the surface without risk to damage the layer. My compounds give better result in preparative TLC. I use Silica Gel G from merck containing gypsum as binder. A bright white spot indicates the desired compound and the entire plate gets yellow colored, even the spot of the reactant is clearly distinguishable. However, it takes some time in the Iodine chamber.

Some of my friends working on something similar but with aromatic moieties have suggested me to use permanent silica gel plates and develop it by spraying vanillin solution. However, I couldnt understand the chemistry behind it. Although I'm still managing the reaction. The compounds under study are peroxides, cyclic and open chain, bishydroperoxides, hydroxyhydroperoxides and tetraoxanes.

Have you tried potassium iodide water solution (slightly acidified with CH3COOH) as spraying agent? In combination with water-soluble starch, this is full analog of peroxide indication strips.

Some exact recipes for peroxide visualization spray:

1) NH4SCN (0.2g) in 15mL of acetone mix with 4% water solution of FeSO4 just before spraying. Red-brown spot will appear immediately

2) Solution a) from 4% water KI and CH3COOH (1:4) , adjust with 1% of water Na2SO3 to colorless solution

Solution b) - 1% starch

Spray with solution a), then after 5min - with solution b).

Peroxide give blue spot.

3) For stable, non-reducible with KI peroxides - > water solution of iron alum

NH4Fe(SO4)2·12 H2O and KSCN
