The RH blood group system is encoded by RHD and RHCE genes, and the difference of G>C at the 676 base of exon 5 of RHCE gene represents the RHE and RHe alleles. If you want to design primers for RHE gene, how should you design them? The primers cited in the previous literature have poor specificity. Our purpose is to distinguish the RHE from RHe, since the two are the alleles and share much more DNA sequence similarity.
Since others have found ARMS primers hard to design you could look at DCAPS. Adding another mismatched base in the primer can almost alwaus produce a restriction enzyme cut site for each allele thus defining the original sequence depending on whether the enzymes cut the pcr product or not.
