When culturing species of Acidithiobacillus sp., pH of medium was shift during the cell growing. How did we figure out the optimal grow pH of such species. Should we just adjusted initial pH at different level and let it go or maintain the pH at stable value?
It is better that you check pH and OD or colony cunt during the culture to optimizing your culture.
Usually the pH increases during the first days, after culturing. However, it will drop by increasing the bacterial activity and no need to set it in the first few days.
When a bioleaching test is carrying out, if the pH rise to more than 3.5-4 during first days it is better to decrease it by adding H2SO, because the growth of these microorganisms will decrease dramatically at these pHs and they cannot grow to produce H2SO4.
