I have a very general question. Since PBS is a isotonic solution, I was wondering if 2X or 0.5X PBS could be considered as hyper or hypo osmotic conditions respectively? I know that typically people use some sort of non metabolizable sugar such as mannitol to create hyperosmotic condition but what about just increasing the salt consentrations as in case of 2X PBS?

2x and 0.5x PBS are hyper- and hypo-osmotic conditions respectively when we compared with PBS.
You can use 2x or 0.5x PBS but remember that change in salt concentration means more or less positive and negative ions available for various interactions which are involved in keeping the protein together and ionic strength is important factor for protein stability, If you add too much salt, you precipitate the protein out of solution.
We work with porcine semen and we prepare solutioins with sodiium citrate and fructose in distilled water taking 300 mOsm as a reference point (below=hypoosmotis; up=hyperosmotic). Used equipe: Osmometer Vapro Wescor.
