Does anybody have experience with dyes that can be used to stain the cell medium so it appears colored in brightfield microscopy. The dye should leave the cells unstained (i.e. be cell impermeable), it should be applicable to live-cells, not cytotoxic, it should be inert and not immunogenic. Any help would be appreciated.

what about (intra)vital stains / dyes which don't permeate cell membrane (at least) of vital cells (working by 'dye exclusion'): examples: Eosin, Erythrosin, Methylene blue, Neutral Red, selected, untoxic "food dyes" (if such exist), Patent Blue, Trypan Blue, and others.

Concentration of dye (usually low-very low = 0.1% - 0.04%) sufficient but has to be tested for the task.

wolfgang is prefectly right. I only add that Janus Green has been also used to distinguish metabolically active cells from dying cells.
There are - for sure- several other dyes capable of doing the intended job, dear Francesco...(:-)) But for choosing a more sophisticated solution with regard to the question, one needs more detailed information on Erik Müllers (Muellers) task / intended processing of the cell culture (e. g. only one possibility would be to increase the contrast for single - or also floating - unstained cells during microscopical evaluation...
