I have to do in vitro transcription of ssDNA then PCR and using PCR product for transcription by T7 polymerase of repeat containing RNA.So after purification and denaturing gel electrophoresis , how can i check accuracy of transcription and sequence of my repeat containing RNA ?
You can check simply by RT-PCR followed by Sanger sequencing on 3730. Mind you, RT also does not make proofread complements. However, it is so much better than RNA pol.
