I want to study the root architecture of wheat roots using IJ Rhizo attached with a scanner that requires complete removal of soil from the plant roots. What will be the most suitable dispersing agent/ detergent/ cleaning medium?
We use deionized water to wash fine root and some time with tape water depending upon your experiment objectives. keep the samples in the Pretti dishes and slowly removed the soil particles with cosmetic brush and scanned the samples after ensuring the removal of soil particles.

Every factor in your experiment will affect root anatomy (soil type, pot size, plant age, etc.) so you'll want to think about which artifacts you can live with, and which you can't. I often use Turface MVP as my potting medium because it is easy to wash off roots, but I don't know if it would cause artifacts in root architecture that would obscure what you want to study.
