I am growing rice plant (IR64) in both soil and hydroponics and i want to collect the root exudates from soil and hydroponics. As it has been said for a long time that rice is a tough plant to work with therefore I want to know if there is any good protocol has or not which will help me to collect the secretion from rice roots for my study.

Collecting root exudes from hydroponic culture is very easy. You just put the plants in pure water for a certain period of time e.g. 3 hours or so and then process this water to purify root exudates. you might add some antimicrobial agent to stop the degradation of these exudates.

As Khurram Bashir said, you can collect exudates by putting the rice plants into water. If you are interested only in what exudate compounds are present/absent you do not need to worry much about volumes, bathing times and re-uptake of exudates. Things aren't as simple if you wish to determine the (gross) rate of efflux. To determine gross efflux is more complicated because many of the small organic compounds that efflux from roots can also be taken up by roots. One way of determining gross efflux is by blocking active uptake by pre-treating roots with CCCP. Another way is to use isotope labelling, as described in the attached paper
