I’m currently planning to do a small scale compound screening on a kinase of my internet. I have come across a lot of kinase assays in various formats and I’m confused what assay to choose for my initial evaluation of what compounds inhibit my kinase.
Does anyone has a recommendation of how to choose between a coupled assay format vs. substrate phosphorylation assays?
It pretty much depends on the target, available plate readers and cost. Most of the commercial Kinase assays are quite good. Many companies provide kinase-specific kits. If you know your target kinase is commercially available, go for it. Otherwise, try some universal assays like Kinase-Glo®, Transcreener®, etc.
I would still say the gold standard is the classical radioactive filter binding assay that I used to do 15 years back. Nothing is as robust and sensitive as that.
