- Measure the enzyme concentration in the loading solution after completion of the immobilization process, and compare it with the initial concentration. The difference gives the amount of the loaded enzyme.
- Characterize the gel using elemental analysis (CHNSO analysis). This can give the enzyme amount in the gel, in case the gel doesn't content at least N or S elements.
- Use stating of the enzyme loaded gel
- Labile the enzyme with a labeling agent such as Fluorescence Cyanideو then load the gel with the enzyme. Using a florescent microscope will show the presence of the enzyme in the gel.
- Use the enzyme loaded gel to catalyze a proper substrate.
However, choosing a proper method depends on the chemical structure of the gel and the enzyme type.
