At 218 nm the MDEG value of protein is increasing in the presence of ligand , but beta sheet content in bestsel is not increasing. Any other method for this calculation.

Calculate secondary structure content using Circular Dichroism (CD) spectroscopy or X-ray crystallography/NMR, followed by deconvolution with software like DICHROWEB or STRIDE to quantify β-sheet percentage. Alternatively, use bioinformatics tools (e.g., DSSP) on protein 3D structure (PDB file) to assign and quantify β-sheet content.
