I am estimating SOD activity by markland and markland method in which I am not able to understand that how to calculate protein concentration which is a used as the denominator in the estimation formula

You must have done the assay with non amount of protein (usually ug) is your assay. 1 uint SOD is the amount of protein that needed to inhibit reduction of NBT by 50%. suppose you used 100 ug protein in assay and you got 3 sod units you can just multiply to convert per mg protein. hope it helps.

Very simple, just buy a commercial total protein kit and follow the procedure that comes with it to estimate the total protein concentration of your each sample (serum or plasma or homogenized tissue samples).
