I am currently working on stem cell-derived exosomes and need to assess exosomal miRNA relative expression by RT-PCR. can i use the stem cells of origin as the calibrator for calculation of fold change in miRNA expression in the stem cell--derived exosomes (comparative CT method) or will there be another calibrator?

For calculating the miRNA relative expression you would need to normalise using specific small nucleolar RNAs C/D box (SNORDs) if you are using the CT method.
This can be found in miScript SYBR® Green PCR Kit for miRNA detection by real-time PCR using the miScript PCR System.

Thanks sir. I was wondering about the control group needed for calculating the delta delta ct for exosomal miRNA expression. Will it be the exosomes stem cell of origin for example: mesenchymal stem cells will be the control group for MSCs derived exosomes ?
