I am trying to find out a suitable technique to calculate the lipid concentration of LUVs. In between the preparation process I loose a portion of the lipid. So want to know how to calculate the final concentration of the lipids?
If your LUVs are made of lipids with a PC headgroup, there are commercially available kits to quantify them via colorimetric methods, e.g. Phospholipase C Test kit.
I have not done quantification via HPLC-MS, but it should be possible, although I am not sure how the size of the LUVs could affect the result. Also, The LUVs I think will have to be solubilized for HPLC. May I ask which expts you will use the LUVs for? If you need it, for example, for leakage assays, I think the kits would be sufficient enough. Personally, I wouldn't spend so much time optimizing the HPLC protocl, for instance, if I can get roughly the same data for my purpose with a kit. Saves a lot of time and effort.
