Say for example, 2microgram/ml of X(adjuvant) NPs and 1microgram/ml of Y(a nucleoside analogue) giving synergy in invitro conditions in HCT-116 cells, to substantiate the finding in vivo xenografts the how the doses should calculate in mg/kg so that the combination of both the NPs work in a synergistic manner?
How to calculate the similar case in Nanoparticles if given separately(if there are two NPs in two diffferent polymeric system or both drugs encapsulated in a single polymeric system)
As long as the in vivo doses are not toxic, it would be reasonable to give them in the same ratio as the in vitro. This would apply also to nanoparticles whether separately or commonly capsulated. ( unless there is some independent release effect biasing the release of one agent over the other)
C. B. In my opinion, generally the same ratio where you got synergy in In vitro, may not be the same in In vivo. So, I would recommend try different ratio starting from 1:2 (that you got In vitro) and optimize according to responses. As far as dose In vivo is concerned, you may start with the dose of a similar drug used in the same species of animal.
