I want to analyze the chemical composition of bacterial extracted Extracellular Polymeric Substances (EPS) treated with different metal ions in XPS. For that I have extracted the bacterial EPS treated with different metal ions and lyophilized it to make dry powder. As the lyophilized EPS is not completely dry and partly sticky in nature. Can I dissolve it in water (as it is soluble in water) and drop cast over glass slide to analyse using XPS? Please suggest me.
There is nothing wrong about dropcasting for XPS preparation, but I would recommend
a) using a conducting substrate instead of a glass slide, ideally one that contains none of the elements you expect to have in your sample.
b) drying it in a vacuum chamber in which you don't cause a mess if there is a boiling retardation when going towards HV/UHV.
