I want to measure EPSs content in soil (with cyanobacterial-biocrust). Can anyone help me with the template to measure EPS contents (including CEPS and TBEPS) by phenol-sulfuric acid / or anthrone-sulfuric acid method. Thank you so much.
To analyze EPS content in soil with cyanobacterial biocrusts, extract EPS using cation exchange resin or sonication followed by centrifugation, then quantify carbohydrate content using phenol-sulfuric acid or anthrone-sulfuric acid colorimetric assays measured spectrophotometrically against glucose standards. This approach helps differentiate between carbohydrate EPS fractions like CEPS and TBEPS accurately.
