To analyze EPS content in soil with cyanobacterial biocrusts, extract EPS using cation exchange resin or sonication followed by centrifugation, then quantify carbohydrate content using phenol-sulfuric acid or anthrone-sulfuric acid colorimetric assays measured spectrophotometrically against glucose standards. This approach helps differentiate between carbohydrate EPS fractions like CEPS and TBEPS accurately.
