I am a newby on in vivo imaging. We are using IVIS imaging system to study breast cancer metastasis in vivo, but we really do not know how to quantify the images along time. As the time pass, of course, the primary tumor and metastases grow so we must change the adquisition parameters. I would like to know how to properly quantify to able to compare the different time points. Thanks a lot!.
Hi,
I usually use the mode 'auto' to set up the time of exposure, because of course, during the progression of the tumor, the signal will increase. Auto should be the best time for your setting, but it will not change a lot if you already set to some specific time. In general i do 'auto' and '2mn' to try to see the small metastases. (if the primary tumor is too big, i will covert it with some black plastic (you know the one that we used to separate the mice) and it will highlight the small signal as in ovary, bone or brain.
To properly quantify, I do region of interest (ROI), draw a circle around the tumor or metastase and take the 'total' value. (the background is so small value that if your circle is a little bit bigger between the pictures, it will not affect because the background is around 10e3 when you signal should be at least 10e7)
Ensure to be in total flux or radiance, but keep it consistent because you will see a log of difference.
I copy and plaste my value in a excel and see the weekly progression. You can do your curve relative to your 'day of injection' value, or just total signal.
To see the tumor evolution in the picture, you need to keep constant the minimum and max value from the scale.
