Actually I am using an enzyme for first time and it has activity of 1,70,000 U per ml. I have prepared slurry of 200ml volume and want to hydrolyse using this enzyme. Literature reports usage of this enzyme in terms of percentage 0.1-0.5% (Enzyme-substrate). So substrate would be accounted as slurry mass (200ml) or total protein basis as some impurities other than protein would also be present. Secondly, how much amount of liquid enzyme needs to be added to slurry should be calculated ? Is it 0.1 ml for 100ml slurry? How it can be optimised for the reaction?

The percentage you are talking about is either the ratio beween volumes (then it would be 0.1 to 0.5mL of enzyme for 100mL of substrate) or the molar ratio between enzyme and substrate ([E]0/[S]0) in order to respect a large excess of substrate over enzyme. In the latter case you need to know the molar concentration of both enzyme and substrate stocks.
