I am planning to run a DTI study on patients with tumors in the anterior and posterior language areas. Do I need to account for a brain mass in the DTI analysis and if yes - how? If possible, please provide me with references.

What do you mean by acount ? Do you want to calculate the tumoral volum?
You do not need to account for brain mass (e.g. brain size) when calculating the DTI statistics (e.g. FA, eigenvalues, etc). Are you also conducting tractography? The issue is that much of the diffusion signal will be effected by the diffusion signal associated with edema related to the tumour, making it difficult to track the white matter bundles near the edema and tumour. What sort of DTI sequence do you plan on using (e.g. single shell, multi-shell, b-values, RSI etc)?
What type of brain tumors do your patients have? Have the patients had biopsies? DTI would be affected by the type of tumor, whether it is infiltrating or well-circumscribed. If my understanding is correct, tumors infiltrating the white matter have a larger impact on DTI metrics.
