Can anyone tell me how quickly individual cells can take up a drug resulting in an effect? I see alot in the literature where cells are incubated with drugs for up to 24 hours but no specific information re. how quickly a drug such as doxorubicin is taken up by a cell. E.g. would a 1 hour incubation result in cell death. Perhaps this is something I need to assess experimentally but any assistance would be much appreciated as I am unsure if I am searching using the right terminology.
You could see dox uptake into cells even after 30 min., but it also depends on the dox conc in a medium. We usually use 1 or 5 uM. However, the effect on cell death will be later, and EC50 values are time-dependent.
It depends on dox concentration as well as susceptibility of the cells. Concentration >10 uM is highly cytotoxic.

We performed some time-lapse microscopy experiments with three prostatic cell lines treated with doxorubicin. You can see apoptosis and necrosis and define the important time points.
