I just want to gather insights on how much time does your reaction take when cleaving arginine-rich peptides? The number of arginines is around 4 and above, while the size of the peptide is around 20-30 amino acids.
Simon Miguel Lopez Dear Simon, if you want to cleave the peptide from the resin and simultaneously cleave the side-chain protecting groups, you can use 2 hours + 1 hour for each arginine. So if you have 4 arginines, you will need 6 hours of cleavage.
I was working on a 90 min + 30 min for every R hybrid protocol as recommended by CEM Liberty Blue technical bulletin and our in-house method using 90% + TFA cleavage cocktail. Our upper limit is that, if ever it exceeded 4 hours, there is a chance that the peptide might be degraded too, aside from being cleaved by the resin. Nonetheless, I will try to apply your recommendation too so that I would be able to yield enough crude synthetic peptide.
