If I understand correctly you wish to know if you can use pooled plasma from multiple rats to validate your method?
Yes you can, and assuming the samples are not significantly different and the analyte you wish to measure does not differ much between rats , it should pose no problems.
However, in general, the more variation you introduce in your method validation, the better. The validation should reflect the way the method will be used. Therefore during validation it is wise to use multiple different batches of pooled plasma, or single rat plasmas if possible. Be sure to add a validation step where multiple different rats samples are tested and compared.
