I want to transfect cells with dsiRNA. The duplexes are resuspended so the final concentration of the stock is 100 uM. I plan on using 6-well plates (2 mL wells). I'm not confident in calculating the amount of dsiRNA I need. The protocol I’m using is the Lipofectamine ® 2000 by Invitrogen (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/Lipofectamine_2000_Reag_protocol.pdf). It recommends using 14 ug of DNA in a 6-well plate. It also recommends using 30 pmol (~0.6 ug) of siRNA per 1 ug of DNA. So I interpreted using 14ug/0.6ug = 23.33. But now I’m lost. I’m not sure what units I’m left with. I’m not even certain if 23.3 is the value I’m shooting for. I've read other sources that recommend 100 pmol for 6-well plates. So my two questions are 1). How do I calculate the dsiRNA volume I need? 2.) What concentration will I need for a transfection?
