I am working with an unusual bacteria that is very hard to lyse.

I want to extract protein and RNA from this bacteria. I need to determine the optimal lysis conditions for this bacteria, but I am unable to find an assay for measuring lysis.

I have tried:

1. OD600 before/after lysis: The difference was very small between lysed and unlysed

2. A280 and A260 of cleared lysate to measure DNA and protein release: The difference was very small between lysed and unlysed

I cannot easily use a microscope to look at bacterial lysis, as the bacteria is very dangerous.

Does anyone know an easy assay to determine proper bacterial lysis?
