Thinking about getting astrocytes derived from IPSc. So I read that first both astrocytic like cells and neurite like cells grow together and to purify astrocytes 3 passages are made.My question is how many more passages after these can the iPAST have?
You have to avoid them becoming confluent. Split them Whenever they become 80 to 90%. After some time, depending on the level of differentiation, the cells will gradually stop proliferating, after this stage you will not need to pass them. You can of course move them to different plates for assays.
This might be after 3 passages or more depending on your line and the differentiation protocol.
In other words depending on the amount of cells you are starting with you will end up with more or less a certain amount of mature Astrocytes at the end of the process. If you are doing 1:4 splits, 1 well will give you 64 wells after 3 splits.
