let me introduce myself. my name is swara yudhasasmita. i'm a student in Gadjah Mada University Indonesia. i had read this article and i follow the method to my thesis. but i want to ask you. how many chitosan that you can collect in this research if you use just 20 mg chitosan? because i think it is too a little. and then do you use centrifugation for seperating pellet and suspension? how about the speed of centrifugation? thanks

By using 20mg, you can't collect grams as in your question. Definitely you are able to collect few mg-s. The exp section in the paper is not informative. Better contact the authors directly for further information

I think that much of nano-chitosan will only appear as a small pellet (about 1-2 mm) at the bottom of the centrifuge tube. As for the detail about the force and the time needed, I think there are lots of information that you can find on papers about nano-chitosan.
6,000 rpm in 20 minute is enough for separate nanochitosan particles. You should follow another procedure (another paper) in order to get bigger content of Nanoparticles. This thing is easy for you to dry and have enough content for adding to your experiment. There are two ways to make nanochitosan: capping with tpp or with MAA. After freeze drying, the sample look like paparaffin, soft or flacible. You also can dry sample by vacuum drying or even by nomal way at 75 oC. Pay attention on particle size determination (less than 100 nm) before prepare a large scale.
It is difficult to get chitosan nanoparticles by centrifuge. Normally, over 13,000 rpm and depend on the size of the nanoparticles. But, If you use centrifuge, the form of nanoparticles maybe changed. You should use spraying dryer to collect them.

According to this procedure, we get 1.6 g for 1 g of chitosan. For pelletization 12,000 rpm was fixed. Nanochitosan in powder form will appear after freeze drying.
