If you take 10ml blood and separate it on FH density gradient you may get 2-3 million PBMCs per ml i.e.; 20-30 million cells and out of which about 60-75% will be CD3 i.e; around 12-15 millinon . So ultimately you may get around 30-40% of CD3 positive cells as CD8 which will be around 3-4 million.
These number of cells may be enough to get good RNA to do further experiments
