In my lab we normally do cell culture (For karyotyping) from fresh sample, just after collecting in heparinized tubes. Now we are thinking of doing some sample which will come from a longer distance. Its probably takes 10-16 hr to reach in our lab.
Is it possible to do the test with retaining its maximum quality?. If so then, what measures should I need to take during sample transportation?.
So I am asking this question to know about the storage condition that i need to ensure during transportation.
Cytogenetic analysis after 10-16 hr of sampling usually still work pretty well. The heparinized samples should be mixed well by inverting the tubes several times and best be shipped at room temperature.
We have been involved in a long standing clinical study in collaboration with a group in Kiel, Germany who does the cytogenetic analysis. Typically leave behind 2-3 mLs of blood sample in heparinised tubes after taking the rest for our study and ship it the next day reaching them the following day, sometimes even upto 72hr after collection. Have had no problems.
I agree with previous comments. If blood is trasported at normal ourside temperatures (~15-25°C) up to 48 hours are normally no big problem for quality and quantity of evaluable metaphases. The longer transport lasts the less metaphases one gets and also banding resolution goes down. At higher or lower outside temperatures such problems may arise after shorter transportation time. So good thermal isolation of the transport box may help also in your case.

Sir, we normally collect the blood in pre-heparinized syringe. Where, normally we take 20 micro litter of Heparin. So, Is it will be okay for 48hr in 15-25C temp ?
We had experience in culturing the whole blood after one week storage, and it's ok.
Thank you all for yours kind information, hope it will be so helpful for me to carry the sample even in prolong (12hr) time of transportation maintaining the conditions towards a quality testing and analysis.
We use to collect blood and inoculate same day. But I have seen people preserve blood in refrigerator and using that if there is a culture failure. They were using KaryoMAX medium.
I always use fresh whole blood samples to karyotyping of farm animals or human. but in some experiments I had a acceptable metaphase index with blood samples which stored for 3 days in 5 degree of centigrade. But in this case adding a little more blood is recommended.
As suggested Fresh samples are always preferred to do cytogenetics / karo typing, but 15-16 hrs is not much delay and we get good results after 1-2 days of stored samples .
Extra sample(cells) can be put to do culture and do check the storage conditions while carrying samples that will also decide the quality of culture and quality of results ( 4-5 deg cel temp in transportation will keep samples intact for 15-16 hrs)
yup.... if temperature of your city is around 25 to 40 then you should no need to bother for 24 to 36 hr..... you can send it at room temperature. and you can also get the good results after one month if you u preserve at 4 degree centigrade.
If the heparinized tube with blood sample will be transported in 4-8 deg. temperature there is no problem with setting culture and karyotyping. You can also store sample in the fridge for few days, it should work still.
I know someone who is sending with DHL the blood collected in heparinized tubes: 1-2 days is OK. Obviously, it depends which kind of analysis will you perform.
I want to store blood for hormone analysis, but due to covid issue there is curfew here, for how long i can preserve blood in light green caped tubes, and at which conditions, so that i would peform elisa for hormonal analysis later in.
