I am interested in extracting DNA from liver and tails to sequence the genome of an African mammal species. It is extremely difficult to get dry ice to the area where the samples will be obtained and I wanted to know if anyone had a successful DNA extraction from tissue samples at room temp for 2-3 weeks in ~95%etOH?

I have collected mammalian tissue in 96% ethanol in field conditions: we were about a month in a steppe (day temperature +25 - 35 C), only after coming back to the lab I put the samples in +4.
I have not used my DNA for NGS, but I experienced no problems with extraction sufficient amounts of quality DNA for other applications.
I see no problem with your sampling! Just put a bit more ethanol per sample to ensure it penetrates all of the tissue fast.

Thanks for your reply. We did a test run here in the lab and placed 10mg of liver tissue in a 2ml tube with 95% etOH which was refreshed 12, 24 and 48 hours after initial submergence. We left the tube for 1 week at room temp and extracted the DNA. It gave a suitable yield of 8micrograms and was high molecular weight (>1kb). Phew :)
Hopefully this will now work on my more precious samples!
