I work freeze dried culture. I planned 3 steps. First step, freze dried culture thawing in liquid medium.Then inoculation on petri plate. Second step, after thawing spore solution incubate at 2 hours then inoculation on petri plate. Third step, after thawing spore solution incubate overnight then inoculation on petri plate.
I want to ask two questions:
1) Are these steps that I planned right? ıs it just first step right?
2) How long spore solution storage in liquid mediım at refrigerator? (Between three steps).

1- in first step must be re-applied three times.
I working on Bacillus spp. I kept it at 3-6 months in liquid media but in slant method at one year.
I dont know the species you are working with--but if it has endospores--it will grow..the problem may actually be killing it--

It is important to know the type of culture you are working with. Generally bacterial cultures can be stored up to 3-6 months in liquid media at refrigerator while in normal storage conditions. Use of 15% or 30% Glycerol stock is another prerequisite for this purpose.
