Which will be the best vector to clone 3kb insert?
Up to ~5kb may be okay. The smaller the better, and as you try to clone larger fragments, the efficiency decreases.
You would be fine with 3kb insert in any of the pUC series plasmids.
With cloning vectors, including pUC series, you can easily go up to 7kb insert. You still will get enough colonies with chemical competent cells (RbCl2).
