At the lab we've been culturing V. Indigofera (strain: NRRL B-3162), but we can't get it to produce indigoidine. We've tried growing it on TSA, PCA, and LB agar, incubating at 25° C.
Even well separated colonies aren't blue.
Have any of you had this problem?
This might help:
Pseudomonas indigofera Agar
Composition per liter:
Agar ............................................................................................ 12.0g
Yeast extract................................................................................ 10.0g
Glucose ......................................................................................... 5.0g
Sodium acetate.............................................................................. 0.5g
pH 7.0 ± 0.2 at 25°C
Preparation of Medium: Add components to distilled/deionized
water and bring volume to 1.0L. Mix thoroughly. Gently heat and bring
to boiling. Distribute into tubes or flasks. Autoclave for 15 min at 15
psi pressure–121°C. Pour into sterile Petri dishes or leave in tubes.
Use: For the cultivation and maintenance of Pseudomonas indigofera.
Acetate is an important medium component for pigmentation of the bacterium. 0.05% is sufficient. Prolonged incubation is another problem which forces colonies to develop metallic copper sheen in place of the blue pigmentation. Remember it is a strict aerobe and mesophilic. There are several media that facilitate pigmentation, apart from the one suggested by Jakub. Read the attached, please.
I tried with the medium suggested by Jakub, and it grown yellow again. I'll try with the mediums described in that papers, Maurice. If still nothing works, may it be we'd have to assume the bacterium we have is not the one we think.
