I wanted to preserve Klebsiella pneumoniae, Rhodococcus equi and Mycobacterium tuberculosis. What is the best technique to preserve these bacteria for a longer period of time? Also, I wanted to know how long we can preserve them. Please specify preservation conditions as well if possible.

Many options are available for the preservation of bacterial strains. These are Refrigeration, Freezing, Freez drying. The most specific option is selected based on the time period of your preservation.
Suppose, If you want to preserve your sample for 2 days, put your sample in liquid media grow them, till it attent the log phase, then put it in a laboratory refrigerator. If you want to preserve for a week. Repeat the same and put the sample to - 40 freezer. If you want further preservation, you need to go for cryopreservation. Long time preservation of bacterial samples is through lyophilisation. Procedures are available in literature provided the availability of the apparatus the lyophiliser.

Freeze dry/ Lyophilization. For few years, one can preserve under sterile paraffin also.@
