I want to mimic the actual condition of the fungi by placing them in a surface and apply fumigation (concentration x time). Later assess their viability by looking at their germination. I will be working with Alternaria, Botrytis and Cladosporium. If somebody has worked on dry spores please share your thoughts and idea. I have something in my mind but would like to make solid with your experience. Thanks.
I think you can use the NA and PDA plated inoculated with dry fungal spores to check the effect of fumigation. These plates will be exposed to chamber having the fumigation. You can compare the difference in growth with plates not exposed to the fumigation.

Thanks for the answer Vikas Kumar, but don't want them to grow at the time of fumigation. We want to see their mortality after fumigation. So, at this point we are thinking of having spores in some inert material, such as vaseline, to stick spores during fumigation. We will grow spores on PDA and will use dry spores. We will have to quantify how much spores we used in experiment. Here we are not trying to use spore suspension and thus difficulty in quantifying them before we use them in experiment. I will appreciate any suggestions from scientists around the world.

I have been collecting spores on empty 60 mm petri dishes by tapping the back of culture plates. Then exposed to fumigation, added about 500ul of sterile water with Tween 20 and plating them on half PDA. Then incubate plates and observe their germination. I am not concerned at amount of spores added to plates before fumigation. I plate the suspension and observe dead and alive spores, that's it. This has given me good result.
