Please tell me the procedure to store the pancreas without any damage to the cells for the estimation, after 5 or 10 days of the excision of pancreas from rats.
Soon after autopsy rinse the organ with .8% chilled saline and place it into a plastic seal-able bag.usually we store it in -20 C if its for estimation within one week of autopsy. or at -80C if its for estimation within one month or so.
Cut out the desired amount while its still frozen. Don't allow the entire organ to thaw.
From my experience liver's parameters tends to be more stable than that of kidney.
The pancreas is particularly susceptible to degradation largely because of it's high proteolytic enzyme content. In addition to previous post consider including protease inhibitors in your saline and if you are retrieving a large number of pancreases do not wait until you have retrieved them all before moving to -20/-80. You could also consider using chilled UW solution as used in clinical organ transplantation. Not essential but contains quite a few goodies that help tissue preservation.
