How do you store and preserve RNA samples to maintain their integrity and stability over time, and what are some of the best practices for long-term storage?
Some basic practices would be:
-Work with your RNA always using benchtop coolers and RNAase free environments and consumables. Clean surfaces with agents such as RNaseZAP.
-Quantify your RNA.
-If you plan several experiments with the same sample then prepare single-use aliquots to avoid freezing-tawing cycles.
-Your RNA should be eluted in Nuclease-Free Water containing DEPC a potent RNAase inhibitor.
-Store your RNA at -80 °C.
TE buffer can also be used for storage as the slightly alkaline pH reduces hydrolysis and the EDTA chelates any contaminating metal ions.
Freeze at -80C in single-use aliquots, and defrost on ice. Freeze-thaw cycles can cause RNA degradation, particularly if there are contaminants in your RNA samples.
