Please tell me how can I sterilize oil (vegetable oil/lipid substrate) before pouring it into growth medium. Someone suggested not to add oil during autoclaving, but then how can I sterilize it?
I am not an expert in that issue but I don't think oil could resist such a high temperature without losing its properties. Maybe it would be better to sterilize it by filtration (0.22 microm)
I have personally filter sterilized vegetable oil using a syringe and a 0.22 micrometer filter. It is time consuming and required a great deal of force. It comes out quite slowly but it definitely works. I would recommend using a larger syringe such as 35 ml so it doesn't break.
I think oils containing no additives (flavors etc) have no water and oxygen to sustain life, so i think is auto sterilized. What are you trying to measure? If bacteria seeds exist in your oil or if interferences are caused from oil in your growth medium ?
Fats and oils should be sterilized by dry heat. The method is, therefore, more convenient for heat-stable, non-aqueous materials that cannot be sterilized by steam because of its deleterious effects or failure to penetrate. Such materials include, powders, oils, and some oil-based injectables.
Preparations to be sterilized by dry heat are filled in units that are either sealed or temporarily closed for sterilization. To sterilize mineral oil I use a flask with aluminum foil lid (2hs x 160ºC). Good look!
I see a lot of answers have indicated that autoclaving vegetable oil is not the way to go. However, I can say from my own experience that autoclaving plant oils for use in growth media for microbes has worked very well for me and my colleagues. We have sterilized palm oil, palm kernel oil, vegetable oil, and pure trioleate by this method.

Can an instrument made of copper or steel smeared with herbal oil be autoclaved? (To clarify,the query is whether herbal oil smeared instruments cannot be autoclaved in a standard autoclave). grateful for a reply from experts or people with experience
